Detecting DNA of multispecies dinoflagellate cysts in the sediment from three estuaries of Makassar strait and fishing port using CO1 primer: Is it CO1 primer suitable for detecting DNA dinoflagellate?

Most dinoflagellate had a resting cyst in their life cycle. This was developed unfavorable environmental condition. The conventional method for identifying natural sediment requires morphological observation, isolating, germinating and cultivating the cysts. PCR is highly sensitive detecting sediment. aim of this study to examine whether CO1 primer could detect DNA multispecies cysts from our sampling sites. Dinoflagellate extracted 16 samples. using COI running. sequencing BLAST. Results showed that there were two clades four locations study. Clade 1 dominated by samples Jeneberang Estuary (JB), Maros (M) Pangkep Estuary(P), while clade 2 Paotere Port (PP). genetic distance varied between ranging 0.5 -0.6. closest sample JB1 JB2, farthest PP1 PP2. not suitable due only picking one DNA, which diatom (Licmophora sp).
